Abstract:
Messenger RNA (mRNA) was discovered back in 1960s, but only the COVID pandemic brought the need for its large-scale production. Fulfilling those opportunities imposes significant challenges on process developers and column manufacturers to improve existing downstream processes. Focus of Sartorius BIA Separations in recent years was the development and commercialization of set of chromatographic columns and processes covering all areas from purification of raw plasmid DNA (pDNA), through monitoring the IVT reaction up to final mRNA polishing. The core of our technology are monolith chromatographic columns (Convective Interaction Media CIM®), especially suitable for separation of large biomolecules due to the convection-based mass transport of analytes to the active surface. The established chromatographic mRNA toolkit includes: 1) CIM Oligo dT columns for polyadenylated mRNA capture; 2) CIM PrimaS and CIM Swiper for capture of nonpolyadenyalted mRNA, including self-amplifying mRNA; 3) CIM SDVB columns for removal of double stranded RNA impurities; 4) CIM C4 HLD columns for mRNA polishing as well as linear pDNA purification (raw material for IVT reaction). All the columns are available in large-scale as well as in analytical and multiwell formats and their potential and applications will be presented during the workshop.
