This poster describes a two-step purification process for coagulation factor IX (FIX) from undiluted cryoprecipitate plasma using CIMmultus DEAE (capture) and CIMmultus SO₄ (polishing) monolithic columns. The CIMmultus columns enable the rapid processing of large plasma volumes and direct loading without prior dilution, which addresses the limitations of conventional diffusive-based resins.
The DEAE CIMmultus step achieved 49% FIX recovery, while the SO₄ polishing step yielded 93% recovery and a final FIX specific activity of 455 IU/mg protein. Co-purified vitamin K–dependent coagulation factors (FII, FVII, and FX) were reduced to below the limit of quantification in the main CIMmultus SO₄ elution fraction. SDS-PAGE analysis confirmed the high purity of FIX.
