Abstract:
Chromatographic analysis of complex AAV presents challenges due to the presence of various impurities, including proteins, DNA, and other matrix species that may coelute with target molecule. In case where one-dimensional chromatography is insufficient for satisfactory separation, more efficient analytical tools are required to address these issues.
This webinar will explore the determination of percentage of full capsids in complex samples, including lysed harvests and spent media, with rapid analysis (results provided within 25 minutes per sample) and baseline separation of empty and full capsids.
Practical applications of the PATfix AAV Switcher will be highlighted, demonstrating its role in optimizing upstream processes through two rounds of DoE. Additionally, we will discuss how this analytical tool, alongside digital PCR (dPCR), helped narrow the harvesting time window, reducing accumulation of empty capsids. Finally, participants will gain insights into how the novel analytics can be integrated into their workflows to enhance efficiency and product quality.
