This method guide presents a novel approach for reproducible full AAV enrichment. The improved method uses a simple linear gradient with organic modifiers and alkaline earth metal salts, resulting in improved AAV E/F separation. In combination with the QA HR line, it enables seamless transition when scaling up and offers consistency when performing isocratic step elution approach.
