Title: Monolithic Chromatography in Viruses and Viral Vector Applications for Purification and Analysis

Abstract:

Viral vectors, such as adenoviruses, adeno-associated viruses, and retroviruses, are key tools in gene therapy, oncolytic therapy, and vaccines, used in numerous clinical trials. Wild-type viruses like bacteriophages are also gaining importance due to antibiotic-resistant bacteria. Monoliths, highly porous chromatographic media, are ideal for purifying viruses, proteins, and nucleic acids due to their high flow rates, binding capacities, and low shear forces. First described 20 years ago for virus concentration, monoliths are now used in various therapeutic products.

The presentation covers recent applications, including:

  • Purification of lentiviral vectors with over 60% yield and significant impurity removal using anion-exchange monoliths.
  • Removal of 7 logs of endotoxins from bacteriophage lysate with two-step monolithic purification.
  • Simultaneous analysis of Orf-virus concentration and purity using chromatography with a multi-angle light scattering detector (MALS).
  • Contributions of monoliths to gene therapy, including AAV purification with cation-exchange chromatography and separation of different AAV species with anion-exchange chromatography.

About the event

The inaugural Power of Viruses conference, held in Poreč in 2018, comprehensively covered key topics in virology. This time we will gather experts from Croatia and abroad to discuss numerous issues related to viruses, exchange experiences and knowledge, stimulate further development of virology in Europe, particularly in southeastern Europe, and promote international collaboration among virologists across different sub-fields including medicine, plant, and veterinary virology.

The scientific programme will be prepared by an international scientific committee of virologists from various fields.
