Optimization of AAV Downstream Process Development from Harvest to Final Product

Abstract:
When it comes to purification of Adeno-associated virus (AAV) achieving a platform process is rather difficult due to the inherent heterogeneity of AAV particles, the variability introduced during upstream processing (USP) and the multitude of AAV serotypes, each with unique properties and therapeutic potentials. The absence of a one-size-fits-all platform process demands a tailored approach to downstream processing (DSP) for each AAV serotype and production condition.
This presentation will cover considerations and examples of case studies for various AAV serotypes for all DSP steps. We will present data from initial lysate clarification and pre-capture options to reduce contaminants, comparison of affinity and ion-exchange capture step and finally optimization of polishing (full enrichment) step on various monolithic columns, all supported with orthogonal analytics.
