Immobilized enzyme reactors (IMERs) on monolithic supports were developed for efficient plasmid DNA linearization, a critical step in mRNA production. EcoRI, NdeI, and HindIII were successfully immobilized, achieving comparable linearization efficiency to in-solution methods without the need for enzyme removal. HindIII IMER-linearized pDNA was directly used in IVT reactions, confirming its suitability for mRNA synthesis.
