The AAV Switcher Platform is a next-generation comprehensive solution for USP monitoring, designed for the efficient enrichment and separation of AAV vectors. Platform Configuration
Step 1: CEX Capture Column (Cation Exchange)
- Utilizes a capture step tailored for robust AAV capsid binding.
- Facilitates initial enrichment of AAVs and removal of bulk impurities, including host cell proteins and DNA.
Step 2: AEX Column (Anion Exchange)
- Positioned downstream in tandem with the capture step.
- Enables high-resolution separation of AAV8 subspecies, particularly effective in isolating full capsids from empty or intermediate forms.
- Exploits charge-based differences in the capsid population for improved analysis.
