The AAV Switcher Platform is a next-generation comprehensive solution for USP monitoring, designed for the efficient enrichment and separation of AAV vectors. It leverages a tandem 2D chromatography setup, which is featured to distinguish between full capsids and other subspecies, such as empty or partially filled capsids.

Platform Configuration

Step 1: CEX Capture Column (Cation Exchange)

  • Utilizes a capture step tailored for robust AAV capsid binding.
  • Facilitates initial enrichment of AAVs and removal of bulk impurities, including host cell proteins and DNA.

Step 2: AEX Column (Anion Exchange)

  • Positioned downstream in tandem with the capture step.
  • Enables high-resolution separation of AAV8 subspecies, particularly effective in isolating full capsids from empty or intermediate forms.
  • Exploits charge-based differences in the capsid population for improved analysis.
