Monoliths offer capacities similar to porous particle columns for proteins of about 1 MDa (~20 nm) but with flow rates 20–50 times higher than columns, they offer much higher productivity. In addition, and very important, flow rate is not a process variable in monoliths. Capacity and separation performance remain constant no matter what the flow rate. Shear stress is an issue, as with other large biomolecules, and the lack of turbulent shear in monoliths can be a significant factor in recovery and long term stability.
